- W2062827626 abstract "Deregulated expression of Notch molecules is observed in many malignant tumors, however, the expression of Notch1 and Jagged1 in colon adenocarcinoma is still unknown. This study is to investigate the expression of Notch1 and Jagged1 in human colon adenocarcinoma. Sixty-five human colon adenocarcinoma and 60 adjacent nontumor colon tissue sections were detected by immunohistochemistry. Ten paired fresh surgical human colon adenocarcinoma and adjacent nontumor colon samples were analyzed by Western blot and RT-PCR. Both Notch1 and Jagged1 were expressed in the cytoplasm of neoplastic cells of colon adenocarcinoma tissue. The protein and mRNA levels of both molecules were higher in colon adenocarcinoma than in adjacent nontumor tissue. Moreover, Notch1 was positively correlated with tumor stage. This investigation demonstrates that Notch1 and Jagged1 are up-regulated in human colon adenocarcinoma and suggests that Notch1/Jagged1 signaling might play a role in the development of colon adenocarcinoma. L’expression anormale des molécules de Notch est observée dans beaucoup de tumeurs malignes. Mais l’expression de Notch1 et Jagged1 dans l’adénocarcinome du côlon est encore inconnue. Cette étude est destinée à étudier l’expression de Notch1 et Jagged1 dans l’adénocarcinome du côlon humain. Soixante-cinq adénocarcinomes du côlon humain et 60 coupes de tissus non tumoraux à côté du côlon ont été détectées par immunohistochimie. Dix paires d’adénocarcinome chirurgical frais du côlon humain et les échantillons à côté du côlon non tumoraux ont été analysés par Western blot et RT-PCR. Les résultats ont montré que des molécules Notch1 et Jagged1 sont exprimées dans le cytoplasme des cellules néoplasiques des tissus d’adénocarcinome du côlon. La protéine et les niveaux d’ARNm de ces deux molécules ont été plus élevés dans l’adénocarcinome du côlon que dans les tissus non tumoraux à côté. En outre, Notch1 est positivement corrélé avec le stade tumoral. Cette étude démontre que Notch1 et Jagged1 subissent une régulation vers le haut dans l’adénocarcinome du côlon humain et suggère que Notch1/Jagged1 signalisation pourrait jouer un rôle dans le développement de l’adénocarcinome du côlon." @default.
