- W2063215468 abstract "RATIONALE: Ara h 2 and Ara h 6 (Ah2/6) have been identified as the major contributors to peanut effector activity in an in vitro degranulation assay. This study utilized a mouse model to assess the in vivo importance of these peanut allergens. METHODS: Soluble peanut proteins were extracted from whole seeds and depleted of Ah2/6 by gel filtration chromatography or via Ah2/6-specific anti-peptide antibodies attached to agarose. Mice were sensitized to peanut via oral gavage, and then underwent two separate challenges in which reaction severity was assessed by measuring body temperature, symptom scores, and mouse mast cell protease (MMCP-1) release. Mice were challenged with recombined fractions following gel filtration (Frac(+)Ah2/6) or recombined fractions excluding the Ah2/6-containing fraction (Frac(-)Ah2/6). Another challenge was performed intravenously with peanut extract passed over a column with pre-immune IgG (control) or immuno-depleted of Ah2/6. RESULTS: Mice challenged with Frac(+)Ah2/6 experienced body temperature decreases of 3.8±0.5°C (mean±SD) while mice challenged with Frac(-)Ah2/6 experienced a significantly lower decrease of 0.9±0.8°C (p<0.001). Symptom scores were significantly less for mice challenged with Frac(-)Ah2/6 (p<0.05). In the second challenge, mice given the control extract had a temperature of 33.7±1.0°C, while the immuno-depleted group was at 36.0±1.7°C (p< 0.05). Symptom scores and MMCP-1 levels were also significantly lower for the immuno-depleted group (p<0.05). CONCLUSIONS: Ah2/6 are major contributors to anaphylactic reactions in vivo." @default.
