- W2063308695 abstract "High-resolution autoradiographs were made of ultrathin sections of the archegoniate regions of gametophytes of the fern Pteridium aquilinum fed with tritiated thymidine. The radiographs of the somatic cells showed that most of the radioactivity was confined to the nucleus. A little was present in the cytoplasm, principally in association with chloroplasts. The radiographs of the egg cells were strikingly different. There was considerable radioactivity in the cytoplasm; according to the grain counts, even more than in the nucleus. These results are in general conformity with those obtained earlier by conventional autoradiography (Bell, 1960), when stringent enzyme experiments confirmed that the radioactivity, both intra- and extra-nuclear, indicated DNA. The high-resolution autoradiographs showed that the extranuclear radioactivity of the mature egg was principally in the mitochondria and proplastids which are present in large numbers in the dense cytoplasm. It is consequently concluded that these organelles contain DNA, in conformity with evidence from other sources. The amounts of radioactivity in the mitochondria and proplastids of the egg were surprisingly large. It is suggested that in this unique cell these organelles may contain more DNA than elsewhere in the life-cycle. This would facilitate their rapid multiplication by fission in the early stages of embryogenesis, independent of DNA synthesis." @default.
