- W2063669673 abstract "The application of the alkaline diaminobenzidine technique for visualization of catalase to mouse heart revealed numerous dark brown particles in the sarcoplasm of myocardial fibers. By electron microscopy the reaction product was localized in 0.2–0.5 μm particles which were located mostly at the junction of A- and I- bands and in close association with mitochondria and the sarcoplasmic reticulum. In control preparations the particles had the typical ultrastructural characteristics of microbodies with a finely granular matrix surrounded by a single limiting membrane but lacked crystalline nucleoids. In biochemical studies it was shown that the highest specific activities of catalase, d-amino acid oxidase and l-α-hydroxy acid oxidase were found in association with the microsomal fraction which contained numerous catalase-positive particles similar to those demonstrated in tissue sections. These findings suggest that the catalase positive particles of myocardium contain in addition to catalase two flavin oxidases and are thus identified morphologically as well as biochemically as peroxisomes." @default.
