Many histochemical processes before (fixation, stabilization, clearing and impregnation, slide-drying, and re-exposing of the antigens) and during (incubations of the antibodies) the immunogold-silverstaining can be accelerated by means of microwave irradiation. To achieve highly reproducible results with respect to labeling intensity and tissue morphology, close attention has to be paid to the constancy of: conductivity of the medium, induced kinetic movement, radiation level, size of the object in relation to the radiation, dilution and ionic strength of the medium, the placeofthe (additional) load in the microwave oven, and differences among microwave ovens.Microwave irradiation, before and during the irnmunogoldsilverstaining (IGSS), can result in an excellent morphology and a very strong IGSS of antigens. In some cases the concentration of the antibody solutions can be lowered, which makes the IGSS procedure cheaper. Combining the antigen retrieval system with microwave irradiation sensitizes the procedure, making it possible to detect antigens which normally cannot be stained.
