The individual components of the dinucleotide fractions of 28-S rRNAs of kidney and adult and embryonic liver of rats, and AH-130 ascites tumor cells were compared after digestion by T1 ribonuclease. The results suggested that rRNA of kidney and adult liver are similar but might differ from those of rather rapidly proliferating tissues, such as fetal liver and AH-130 tumor. 2. The nucleotide compositions of nucleolar 45-S RNAs of tumor and fetal liver cells differed from that of adult rat liver. 3. Purified rRNAs of polysomes from AH-130 tumor and adult liver cells were digested with pancreatic ribonuclease and the resulting oligonucleotides were separated according to chain length. The isotope content of dinucleotides of RNA of tumor cells was higher, and that of pentanucleotides was lower than in the RNA of adult liver. No difference in the extents of methylation of rRNA in these tissues was observed after alkaline digestion, so the different contents of dinucleotides in tumor and liver cells were not due to differences in methylation of rRNA. 4.
