• W2065657614 abstract "Studies of the mechanism of lymphocyte activation by phytohaemagglutinin have suggested a simple theory of immunity. Lymphocytes, each capable of forming a different antibody, are released from the thymus into the circulating lymphocyte pool. Each cell bears at least two, adjacent, identical, univalent antibody sites at its surface. Reaction of “self” antigenic determinants with both of the cell-borne sites results in complement fixation and the destruction of the cell. Cells not bearing antibody sites of a specificity corresponding to “self” determinants then begin secreting natural antibody (IgM). This antibody is of the same specificity as the cell-borne antibody and buffers the cell against reaction with foreign antigenic determinants. Reaction of an antigenic determinant with one cell-borne antibody site causes the cell to enlarge, secrete more IgM and divide. Two alternative processes may then follow. If the extra IgM secreted is not sufficient to neutralize all remaining free determinants diffusing into the region, the cell will be stimulated to enlarge again and secrete IgG. If the extra IgM secreted is sufficient to neutralize all remaining free determinants diffusing into the region, post-mitotics will not enlarge and secrete IgG. They may migrate from the circulation into tissue sites of antigen administration and there react with determinants to initiate a “cell-mediated” response. IgG, but not IgM, can leave the circulation and neutralize free determinants located in tissue sites of antigen administration.
