- W2067459680 abstract "100 Aims: The expression of HLA class I and II antigens in human enterocytes is induced by exposure to gliadin. In previous studies, the expression of HLA class I and II antigens in human epithelial cells has been restricted to the basolateral membrane. In the following study, we wished to investigate the time-dependent intracellular staining patterns of HLA-I and HLA-DR antigens in celiac disease (CD) enterocytes after exposure to gliadin. Methods: Duodenal biopsies from CD patients (according to the criteria of ESPGHAN) under gluten free diet were harvested endoscopically, incubated for 5, 20, 60, and 360 min with 1 mg/ml Frazer's Fraction III, and fixated at room temperature in 5 % paraformaldehyde. Ultrathin frozen sections of the duodenal biopsies were labeled by HLA-I (A-B-C) and HLA-II (DR) antibodies. Their binding sites were visualized in the electron microscope by immunogold particles. Results: The labeling intensities of HLA-I and HLA-DR were low at 0 minutes and restricted to the basolateral membrane. There was a marked increase of the labeling density at the subsequent time points. Both HLA antigens were detected in subapical endocytic vacuoles, Golgi stacks and on the basolateral membrane. In contrast to previous studies, we found a significant staining on the apical membrane of CD enterocytes. Conclusions: The rapid increase in HLA class I and II antigens and their detection within the Golgi stacks in CD enterocytes following exposure to gliadin points to an immediate upregulation of HLA expression. Incubation with gliadin also induces that HLA class I and II antigens are not only sorted to the basolateral surface but also to the apical membrane of CD enterocytes. We speculate that HLA antigens present on the apical membrane of CD enterocytes are involved in the uptake of gliadin." @default.
