- W2067570036 abstract "Ribosomal RNA, which was isolated from yeast with Duponal at 100° and had a s20,w of 4.5, was adsorbed to Ecteola cellulose and was eluted with 2 M NaCl at pH 9.7. Amino acid transfer RNA with a s20,w of 3.2 was eluted with neutral NaCl. When chromatographed at 60 and 80°, the ribosomal RNA had a decreased affinity and the transfer RNA an increased affinity for the adsorbent. This datum supports the role of secondary structure in the determination of chromatographic characteristics. When high-molecular-weight ribosomal RNA isolated by the phenol procedure was chromatographed on Ecteola adsorbent, a variable per cent was not adsorbed and retained its high average sedimentation coefficient. The major portion of the adsorbed material was eluted with 2 M NaCl at pH 9.7 and had an average sedimentation coefficient of 7–8. This evidence indicates that the ribosomal RNA was degraded during the chromatographic procedure. The degradation of the RNA was thought to be due to interaction with the Ecteola rather than to the alkaline conditions of elution." @default.
