- W2067861747 abstract "Abstract Conditions are described for use with columns of methylated serum albumin-kieselguhr which permit resolution of bacterial rRNA from its precursors. The key factors influencing the fractionation are temperature and ionic strength. When total RNA from a relaxed control strain of Escherichia coli is fractionated on a column (2 cm × 23.5 cm) with a linear NaCl gradient from 0.65 to 1.06 M (990 ml 0.025 M Tris-acetate, pH 6.5, 27°), three major components of rRNA are separated; these are the 17-S, 16-S and 23-S components. The 22.5-S precursor of 23-S RNA usually appears as a shoulder on the leading edge of the 23-S RNA. These components have been characterized in terms of radioactive labeling in vivo during the growth and starvation of this mutant and also by acrylamide gel electrophoresis. Short term incorporation of radioactive uracil appears exclusively in the 17-S, 22.5-S and mRNA fractions after chromatography. 17-S and 22.5-S rRNA's are synthesized at different rates and, during starvation, accumulate to a different extent." @default.
