The samples are labeled with two spectrally resolvable fluorescent dyes, such as propyl-Cy3 and methyl-Cy5, mixed, separated in a single 2D gel and detected at two different excitation and emission wavelengths. Alternatively, stable isotopes can be used for differential labeling of the control and treated samples. matrix-assisted laser desorption ionization time-of-flight mass spectrometry is used to determine the exact mass of peptides or proteins. Coupled liquid chromatography and tandem mass spectrometry is used for direct analysis of a complex protein mixture without the 2-DE step." @default.
