- W2068961830 abstract "Metabolic syndrome and its components, such as systemic hypertension and peripheral insulin resistance are known risk factors for the development of Alzheimer's disease. Using an animal model of metabolic syndrome, stroke-prone spontaneously hypertensive rats (SHR-SP) fed a high-fat diet, we analyzed the effect of systemic hypertension and peripheral insulin resistance on the hippocampal neurogenesis, a sensitive indicator of CNS stress. At the age of 4 weeks, a group SHR-SP rats was placed on a high-fat diet. Another group of SHR-SP rats, and a group of Wistar-Kyoto rats, both fed the standard rat chow, were used as controls. At the age of 12 weeks, neurogenesis in the dentate gyrus of the hippocampus has been assessed by counting the number of Tbr2-positive and doublecortin-positive neurons. By the age of 12 weeks, both SHR-PS fed a high-fat diet and the group of SHR-SP rat on the standard chow had developed increased peripheral insulin resistance, compared to the group of Wistar-Kyoto rats, as indexed by the fasting glucose to insulin ratio. This effect was more pronounced in the group of SHR-SP rats fed a high-fat diet. Compared to the Wistar-Kyoto rats, SHR-SP rats had increased numbers of Tbr2- positive neurons (p < 0.02) and doblecortin-positive neurons (p <0.02) in the dentate gyrus; however, there was no effect of high-fat diet in the SHR-SP rats. These data suggest that both neurogenesis and early survival of hippocampal neurons were increased in the stroke prone spontaneously hypertensive rat. It is possible that increased hippocampal neurogenesis served as a compensatory response to the CNS stress caused by hypertension and increased peripheral insulin resistance." @default.
