- W206917103 abstract "This chapter focuses on the interactions between MCAT elements and the TEF-1 family of transcriptional regulatory proteins, with particular emphasis on the roles of these components in cardiac-specific transcription. MCAT elements are responsible for diverse transcriptional regulatory responses of both muscle-specific and non-muscle-specific promoters. Two classes of MCAT transcriptional regulatory elements can be distinguished. One class of MCAT elements governs gene expression with a high level of cell and tissue specificity as exemplified by the cTnT gene promoter. A second class of MCAT-dependent genes, exemplified by the SV40 promoter, exhibits a low level of cell and tissue specificity. The TEF-1 genes govern gene activity in a wide variety of plants and animals. There are at least four vertebrate TEF-1 genes, some of which are expressed in a tissue-restricted fashion, with alternative splicing adding to the complexity of expressed TEF-1 proteins. These proteins are likely to bind differentially to different MCAT elements leading to distinct spatiotemporal patterns of transactivation, dependent on the cognate TEF-1 isoprotein. Recent work shows that additional DNA binding proteins are required for TEF-1 family members to act on transcription. In the case of the cardiac troponin T gene promoter, a second, unknown protein appears to bind cooperatively to flanking sequences and to the adjacent TEF-1 moiety bound to the core motif. Since NTEF-1 predominates in nonmuscle tissue, discussion hypothesized that the X-NTEF-1 complex is repressive. By contrast, in muscle tissues where RTEF-1 is abundant, the XRTEF- 1 complex stimulates transcription. The diversity of flanking sequences present in the MCAT elements of muscle promoters suggests that many different proteins may be responsible for modulating MCAT dependent promoter activity in skeletal and cardiac muscle cells." @default.
