- W2069316316 abstract "Abstract Sera commonly used for tissue culture are rich in glycosidases. Horse, calf and fetal calf sera contain one, two and five activities, respectively, which hydrolyse the appropriate p-nitrophenylglycosides at rates greater than 1 nmole/h/mg protein. As these enzymes could potentially degrade glycoproteins and glycolipids which might be related to growth or differentiation, we eliminated them from the serum by treatment at 62 °C. The ability of heated sera to support cell growth and differentiation was tested with a number of cell lines and with primary muscle cells, and was found to be largely unaltered. Normal and transformed mouse fibroblasts were not further affected by growth in the presence of different monosaccharides and glycosides which further inhibit the low levels of glycosidase activity resistant to heating. We conclude that cell growth, growth control and myoblast differentiation are unrelated to the levels of these enzymes." @default.
