- W2069965675 abstract "PURPOSE We tested whether exercise training could improve the Ca2+ dependence of LV performance during acidosis in the spontaneously hypertensive rat (SHR) model. METHODS Twenty-four 4-month old SHR females were randomly assigned to a sedentary (SED) or exercise-trained (TRD) group at (20 m/min, 60min/day, 5 days/week, 6 months), while Wistar-kyoto rats (n =12), served as sedentary controls. Blood pressure and M-mode echocardiography were performed prior to sacrifice. In vitro isovolumic Langendorff performance was established (paced at 5 Hz, LVEDP set at 11mmHg, coronary flow =16 ml/min, buffer-perfused, 2 mM Ca2+). LV performance was measured during:the LV pressure-volume relationship (P-Vrel), and with incremental Ca2+ infusions (2.5 to 4 mM/L) under two pH conditions (pH=7.4 and pH=6.8 -hypercarbia induced). RESULTS Mean blood pressure and HR were elevated in SHR and reduced with training. Heart weight and LV anterior wall thickness (diastole) were increased in SHR vs WKY (P < 0.01) and augmented with training (WKY,1.59±0.03 mm; SED,1.93±0.08 mm; TRD,2.18±0.08 mm; P < 0.05). The P-Vrel was shifted rightward in TRD (P < 0.05). +dP/Dt at pH 7.4 was higher in SHR compared to WKY (WKY, 5433±232 mmHg/sec; SED, 6469±413; TRD mmHg/sec, 6520±399 mmHg/sec); in acidosis, +dP/Dt was better maintained in TRD than WKY (P < 0.01) and SED (P < 0.05), (WKY,2609±277mmHg/sec; SED, 2732±321mmHg/sec; TRD,4250±666 mmHg/sec). The [Ca2+] required to elicit ½ of the maximal LV Dev P during pH 7.4 was not different between groups; in acidosis, TRD required a lesser [Ca2+] to elicit ½ of the maximal response compared to WKY (P < 0.05) and tended to be lower than SED. CONCLUSION These data suggest that exercise training in SHR improves Ca2+ responsiveness such that LV performance is improved during acidosis. Interestingly, LV performance during acidosis is slightly improved in SHR." @default.
