- W2070551004 abstract "P1131 Aims: Inflammation, epithelial cell injury, and development of fibrosis and small airway obliteration are the key features in the pathogenesis of post-transplant obliterative airway disease (OAD). Platelet-derived growth factor (PDGF) promotes fibroblast proliferation and mediates inflammatory responses. We designed this study to assess the effect of PDGF receptor inhibition in a large animal model of OAD. Methods: Small bronchi of 1,5 cm in length were transplanted subcutaneously into 12 recipient pigs. Three groups were formed: autografts, nontreated allografts, and allografts treated with oral imatinib, 10 mg/kg/day, a selective PDGF receptor tyrosine kinase inhibitor. Grafts were harvested on post-transplant days 4, 7, 11, and 14 for histology (H&E) and immunohistochemistry. Results: No signs of OAD occurred in autograft controls. In both allograft groups epithelial destruction was followed by gradual obliteration of the bronchial lumen. In the treated allografts these histological alterations proceeded slightly slower than in the nontreated ones. Inhibition of PDGF receptor reduced inflammatory cell infiltrates both in the bronchial wall and in the pericartilaginous area (p<0.01 on day 11). It decreased the recruitment of CD4+ and CD8+ lymphocytes (p<0.01 from day 7 onwards) and of macrophages (p<0.05 from day 4 onwards). PDGF receptor alpha expression peaked earlier in the nontreated allografts than in the treated ones, while PDGF receptor beta expression was similar in both groups on early timepoints. Conclusions: Imatinib, as single therapy, reduced inflammatory cell recruitment into the grafts. These results further confirm that PDGF has a role in the development of post-transplant OAD, and PDGF receptor might be a future target in OAD prevention." @default.
