- W2072115174 abstract "Membrane-bound (Ca2+ + Mg2+)-ATPase (ATP phosphohydrolase, EC 3.6.1.3), deficient in or saturated with calmodulin, was prepared from human erythrocytes. The dependences on ATP, calcium ions, and potassium ions of the membrane-bound ATPase were studied and compared with the behavior of ATPase solubilized by treatment with detergent (Triton X-100). The Ca2+-ATPase was stimulated by millimolar ATP concentrations, especially the calmodulin-saturated ATPase at high Ca2+ concentrations. The solubilization increased the effect of high ATP concentration, also on the calmodulin-deficient ATPase at lower Ca2+ concentrations. The solubilization strongly increased the low apparent Ca2+ affinity of calmodulin-deficient ATPase, indicating the existence of Ca2+-binding sites on the enzyme distinct from the Ca2+ sites of the dissociating calmodulin. Further, solubilization reduced the modulating effect of calmodulin-binding on K+ activation of the Ca2+-ATPase. The activation by detergent differs from that of calmodulin in several ways. For instance, the detergent seems to abolish the cooperative interactions between enzyme effectors both in presence and absence of calmodulin. The experiments suggest that the binding of calmodulin to the membrane-bound Ca2+-ATPase may change the enzyme from a closed or tense to a more open or relaxed conformation." @default.
