The 16S rRNA sequence derived from the DNA sequence contains 1504 bases and exhibits the characteristic features of the primary and secondary structure of other archaebacterial 16S rRNA's. Helical regions of the secondary structure are extremely rich in G-C pairs, possibly a selected structural property. — The sequence was aligned to the sequences from Halobacterium volcanii, Methanococcus vannielii, Methanobacterium formicicum, and Escherichia coli; the sequence homologies and their conversion into an unrooted tree allow the following major conclusions: (i) The phylogenetic distance between Thermoproteus and the methanogens almost equals that between the halophiles and the methanogens; (ii) the relationship to the sequence of an “outside” organism, Escherichia coli, indicates that the root of the tree may be located in the Thermoproteus branch. If this is supported by additional sequences it furthers the conclusion that all archaebacteria are derived from a common ancestor; (iii) the evolutionary rates of the archaebacterial 16S rRNA sequences differ greatly: Methanobacterium formicicum exhibits the lowest genetic drift; it is somewhat higher in Methanococcus and Thermoproteus and drastically higher in the case of the halophile. — The single operon present on the Thermoproteus chromosome contains no tRNA gene in the 16S/23S rRNA intercistronic spacer; the RNA derived from the spacer can form a “prokaryotic loop” with sequences flanking the 16S rRNA at the 5′ side." @default.
