We used cDNA GEArray to analyze differential expression of inflammatory genes in alveolar macrophages, which may be attributed to MCT-induced disease. Most of the genes regulated in response to MCT were consistent with the M2 phenotype of alternatively activated macrophages, suggesting their role in pulmonary fibrosis. A diet high in retinoic acid (RA) prevented MCT-induced monocyte/macrophage infiltration and severe lung injury. At the transcriptional level, RA reprogrammed the inflammatory expression patterns induced in macrophages by MCT. The identified sets of RA-regulated genes associate alveolar macrophages with classical M1 activation phenotype. Collectively, our GEArray data provide new insights into regulatory role of RA nutritional status in the transcriptional modulation of inflammatory responses in alveolar macrophages, and improve our understanding of cytokine/chemokine imbalance in the pathogenesis of pulmonary fibrosis." @default.
