- W2073246603 abstract "Digoxin was administered iv to healthy beagle dogs at dose levels previously shown to induce sustained toxicosis and to permit survival over a 2-week period. Treatment regimens of both 5 and 7 days/week were employed. Electrocardiographic (ECG) responses, clinical pathology, and serum digoxin concentrations were monitored, and, at necropsy, tissues were examined for pathological changes. The development of digoxin toxicosis among treated dogs was extremely variable and did not correlate well with the amount of digoxin administered. However, correlation between the degree of toxicity and serum digoxin concentrations appeared to be good. The results indicate that of the variables monitored at the sampling times employed, ECG responses were the most sensitive, prompt, and consistent indicators of digoxin toxicity when moderately toxic serum digoxin concentrations (2.5–6.0 ng/ml) were attained in normal beagle dogs. Elevations in activities of serum creatinine phosphokinase (CPK) and lactic dehydrogenase (LDH) were less reliable. At severely toxic serum digoxin concentrations (above 6.0 ng/ml), elevated CPK and LDH activities appeared early and persisted for several days until the mean serum digoxin concentration fell below 6.0 ng/ml. An unexpected finding was evidence of compromised renal function as indicated by elevated serum creatinine and blood nitrogen concentrations and by alterations in serum electrolytes.
