Thls may be a reason for the very complex regulatlon of the amount of thyrold hormone ultlmately acting upon the cells of indlvldual tlssues. Thls complexlty may be necessary to render possible the dlfferentlal regulation of different tlssues and for the detection and meetlng of varlations in demands due to environmental alteratlon, such as changes in food consumptlon, dlsease, etc. In additlon, such a system has a hlgh degree of versatlllty so that defects at one level can be compensated by alteratlons at other levels. In the thyroid gland the complexlty is expressed by a unlque type of hormone synthesls and secretlon (5-7): flrst a big preprohormone, uniodlnated thyroglobulin is syntheslzed in the folllcular cells. Important steps of modulatlon of thyroglobulln occur wlth formatlon of the iodothyronlnes in the peptlde chalns in a speclal extracellular compartment, the collold, whlch also has a hlgh deposltlng capaclty for the formed prohormone. Stimulatlon of the secretory processes leads to relntake of the into the cells for processlng by hydrolysls, which ylelds thyrold hormones. One of the llberated thyrold hormones, thyroxine (T4) , may also be consldered to be a prohormone whlch in part is further processed in the thyrold as well as in perlpheral tlssues by delodination in the outer rlng to the active 3,5,3-trllodothyronine (T~) or In the inner ring to the metabollcally inactive 3~3,5-trliodothyronlne (rT3)." @default.
