The nature of the clinically used photosensitizer HpD is first presented along with structures of molecules found to be efficient in vitro. The possible role of pH in the preferential retention of dicarboxylic porphyrins by tumors is discussed in light of results obtained with membrane models. The uptake of dicarboxylic porphyrins by cells most likely involves passive mechanisms. Cell photoinactivation using a purified porphyrin does not depend upon the incubation time but only on the intracellular concentration of the dye. This likely reflects a poor specificity of the photoinactivation processes with regard to the cellular localization of the dye. The properties which should be presented by more efficient photosensitizers are discussed. Quelques aspects moléculaires de la photochimiothérapie et du photodiagnostic des tumeurs par les porphyrines sont examinés. La nature chimique du dérivé de l'hématoporphyrine (HpD), largement utilisé en essais cliniques, est rappelée ainsi que la structure d'autres photosensibilisateurs efficaces in vitro. L'étude de l'interaction de porphyrines dicarboxyliques avec des modèles de membranes suggère que le pH, localement abaissé dans les tumeurs, est un facteur important dans la rétention préférentielle de ces molécules. L'incorporation de porphyrines dicarboxyliques par des cellules en culture apparaît comme un phénomène passif. La photoinactivation cellulaire par une porphyrine purifiée ne dépend pas des conditions d'incubation mais seulement de la concentration de photosensibilisateur dans la cellule suggérant une relative absence de spécificité par rapport à la localisation du colorant. Les critères que devraient présenter des photosensibilisateurs plus efficaces sont discutés." @default.
