Repeti t ion of this synthesis by two other routes yielded the same final material. 2 All in termediary pept ides as well as the final product gave correct e lementary and amino acid analyses. The schemes of synthesis we had fol lowed excluded any racemization and the action of enzymes proceeded as expected for a peptide of the above mentioned constitution. Nevertheless the synthet ic octapept ide proved to be completely devoid of the typical biological activities of bradykinin. 3. 4, s On April 14, 1960, I communicated our negative results to Dr. Elliott. Following his suggestion, we tested the octapept ide immedia te ly after the last s tep of the synthesis in order to minimize a possible inactivation of the final mater ia l for some unknown reasons and tried also to add several inorganic salts, including magnesium salts, in order to allow for the formation of some possible act ive complexes. But in vain! We also excluded the possibil i ty of the presence of some hypothet ical inhibitors by testing a mixture of the synthet ic octapept ide wi th natural bradykinin. We arr ived therefore to the conclusion that the proposed s t ructure was p robab ly par t ia l ly incorrect. This v iew was suppor ted b y the fact that the behaviour of the synthet ic octapeptide toward chymotryps in differed f rom the one of bradykinin itself. The synthet ic octapept ide was split by chymotryps in s imultaneonsly at the Phe-Ser and at the Phe-Arg bonds, as would be expected for a pept ide of this constitution. On the cont rary, bradykinin had been found by Elliott et al.1 to be much more rapidly split at the Phe-Arg bond than in any other place of the molecule. Dr. Elliott told us that he was puzzled by the fact that the pept ide obtained af ter a short action of chymotryps in on bradykinin yielded only phenyla lanine on the subsequent action of carboxypept idase.
