Materials/Methods: The lung adenocarcinoma cell lines A549 (p53) and H1299 (p53) were utilized for this study. Stable transfections were carried out using pcDNA3.1 carrying MIIP cDNA by Lipofectamine 2000. MIIP expression in selected clones was determined by Western blot analysis. The effect of MIIP on cell proliferation and survival was determined by MTT assay. Cells were irradiated and then assessed by clonogenic assay. Cell cycle alterations were assessed by fluorescence activated cell sorting (FACS) analysis of propidium iodide (PI)-stained cells. The expression of p53 was measured using Western blot analysis. We used nude mice xenografts of A549 cells with stable transfection of MIIP (A549-MIIP) or pcDNA3.1 (A549-pcDNA3.1) and calculated tumor growth delay after radiation to compare radiosensitivity. The study of correlation between MIIP expression and survival of patients are ongoing. Results: Three clones with MIIP overexpression in A549 and two clones in H1299 were selected and used for further analysis. A549-MIIP cells displayed significantly lower mean colony formation rate (p < 0.05) following irradiation at doses between 2 and 8 Gy compared to that of A549-pcDNA3.1 cells or parental cells. In A549-MIIP cells, p53 level was increased and the survival fraction decreased with MTT assay. Significant increases in the percentage of cells arrested in the G2/M phase of the cell cycle were detected in A549-MIIP cells (p < 0.05). Furthermore, irradiation of A549-MIIP xenografts caused significant growth delay compared with A549-pcDNA3.1 in vivo. Conclusions: The results of these studies suggested that MIIP significantly enhanced the radiosensitivity of A549 cells both in vitro and in vivo. It is a promising radiosensitizer for patients with adenocarcinoma of lung. The study of correlation between MIIP expression and survival of patients are ongoing. Author Disclosure: G. Zhu: None. L.
