Mutagenic agents used were: 2-amino-purine (AP), 5-bromodeoxyuridine (BD), sodium nitrite (NO2), t-butyl hydroperoxide (t-butyl) and beta-propiolactone (BPL). Some of the histidine mutants were of spontaneous origin, while others were induced by ultraviolet irradiation (u.v.) and 2-aminopurine. All but one of the twelve auxotrophic derivatives of the LT2 strain, which were originally obtained after treatment with AP, showed an increase in “reverse” mutation frequency with BD. The u.v.-induced auxotrophs gave a variety of responses with the different mutagenic agents and no particular mutational pattern predominated. Of the 54 LT2 strains of spontaneous origin tested, 26% were affected by base analogues, 45% by BPL and 28% by t-butyl. In 52% of the cases, t-butyl and BPL were mutagenic for the same strains. A greater proportion of the LT7 strains of spontaneous origin responded with base analogues and the other chemicals. The increase paralleled the presence of the mutator gene. All chemicals used, except nitrite, were mutagenic with 90% or more of the 80 LT7 strains possessing the mutator gene. The pattern of induction to prototrophy in LT7 strains lacking mutator was very similar to that of LT2 strains." @default.
