- W2075231036 abstract "Instruments are being developed enabling accurate physical measurements to be made on stains, or other deposits, produced within cells by cytochemical staining methods. The theoretical possibility of eliciting information on kinetics of artificial substrate-enzyme reactions in situ, from measurements of rates of stain production, is analysed in this paper. A staining method must conform to a number of strict preliminary conditions before it shows any promise of providing quantitative information. These conditions are much more restrictive than those necessary for the mere provision of good localization of the enzyme. It is unlikely that useful information can be extracted from a process unless the process very rapidly attains a steady state. If the various conditions are fulfilled, and if interaction with the enzyme obeys classical theory, then three cases arise. (i) When penetration of substrate throughout the tissue is very rapid, the Michaelis-Menten equation will apply. (ii) When penetration occurs slowly into a region in which the enzyme is uniformly distributed, a modified Michaelis-Menten rate law will be satisfied under certain conditions. (iii) When slow penetration occurs into a non-uniform enzyme distribution, the problem is intractable.
