- W2075349965 abstract "Abstract The efflux of calcium (Ca) during Ca-free washout of the gas perfused kitten heart is characterized by three monoexponential compartments. One compartment, CaII, is directly related to the maintenance of contractile force. A second compartment, CaIII, is believed to represent Ca arising from tissue binding sites in the cell such as the sarcoplasmic reticulum and the mitochondria which may be involved in Ca sequenstration. Perfusion of hearts with a solution containing 50 μ m lanthanum (La) caused contractile force to decrease immediately to less than 5% of that before treatment. During 30 min of gas perfusion contractile force gradually returned to about 40% of pretreatment levels. In contrast to untreated hearts, Ca-free washout now produced a pronounced contracture. Analysis of the Ca efflux curves showed that CaIII was absent under these conditions. When the Ca-free washout solution contained one-half the normal Na concentration (70 m m ), the intensity of the contracture was greatly diminished, and CaIII again was detected in the washout curve. The results suggest that La treatment may have interfered with the Na activated component of Ca efflux and consequently the Ca:Ca exchange system may then have contributed to net Ca efflux during Ca-free washout. In normal Na concentrations net Ca efflux may have been retarded, but when [Na]o was reduced during Ca-free wash, the ratio [Ca] o [Na] 2 o was increased and hence the affinity of the Ca carriers increased sufficiently to reverse the inhibition of Ca efflux and prevent contracture." @default.
