Both lipids and proteins were covalently labelled upon ultraviolet irradiation. Rhodopsin incorporated the label in regions which are probably burried in the lipid core. Treatment of disc membranes with the water-soluble proteases thermolysin, α-chymotrypsin and pronase failed to hydrolyse significantly the labelled regions of rhodopsin, yet the first two proteases reduced most of the rhodopsin molecules to half their size. The protease-resistant residues retained the carbohydrate moieties of rhodopsin. The results are discussed in terms of the penetration of rhodopsin in the lipid core of the bilayer and its accessibility to carbohydrate and protein hydrolases." @default.
