- W2076067390 abstract "24 Background: Glutamine's (Gln) important role in the immune response has generated interest in its therapeutic use in catabolic states. The use of its dipeptides, Glycylglutamine (GlyGln) or Alanylglutamine, has been recommended due to their favourable pharmacokinetic properties. In measuring lymphocyte proliferation, our objective was to establish whether lymphocytes metabolize Glycylglutamine and Glutamine equally efficiently. This was tested in healthy donors and HIV-infected individuals. Methods: Lymphocytes were obtained from 7 healthy donors and from 11 patients suffering from AIDS. Gln or GlyGln were added to cell cultures at various concentrations between 0.0 mmol/l and 1.0 mmol/l, respectively. Mitogenic stimulation was performed separately by ConA or SAC. Results: The use of either source of glutamine in equimolar concentrations did not result in any overt difference of proliferation. Minor differential effects occurred relating to the mitogen used. Proliferation upon ConA-stimulation was better with GlyGln and peaked at 1.0 mmol/l. When SAC was the mitogen, Gln seemed slightly superior to GlyGln. Optimal proliferative responses were obtained at a concentration of 0.4 mmol/l. A similar response pattern was demonstrated in HIV-patients, even though at lower absolute proliferation level. Conclusion: GlyGln can generally be a substitute for Gln at equimolar in vitro concentrations with regard to lymphocyte proliferation. Differences relating to the mitogen used may reflect different metabolic requirements of the different lymphocytic subsets stimulated. Supplementation of Glycylglutamine may be beneficial for AIDS-patients with regard to lymphoproliferative responses." @default.
