- W2076608509 abstract "Abstract l -Azetidine-2-carboxylic acid, the naturally occurring lower homologue of l -proline, inhibits chlorophyll a and phycocyanin synthesis in cells of the unicellular alga, Cyanidium caldarium. The inhibitory effect of the homologue on pigment synthesis in algal cells was partially reversed during subsequent incubation with proline. Incorporation of radiolabeled proline into algal protein was reduced by 67% and 80% in cells preincubated with 0.4 mM or 1.0 mM l -azetidine-2-carboxylic acid, respectively. However, incorporation of radiolabeled leucine into algal protein was not affected by the homologue at these concentrations. l -Azetidine-2-carboxylic acid stability to hydrolysis in p-toluenesulfonic acid and the methods employed for measurement of l -azetidine-2-carboxylic acid incorporation into protein by amino acid analysis are described. It was shown that radiolabeled l -azetidine-2-carboxylic acid is incorporated into C. caldarium phycocyanin, a biliprotein which constitutes approx. 25% of the soluble protein in fully pigmented algal cells. Amino acid analysis of 14C-labeled phycocyanin revealed that 30%, 21%, and 26% of the recovered radioactivity eluted with the l -azetidine-2-carboxylic acid standard, with methionine, and with serine, respectively. Minor quantities of radioactivity eluted with other amino acids. l -Azetidine-2-carboxylic acid metabolism in algal cells is discussed. It was concluded that l -azetidine-2-carboxylic acid inhibition of chlorophyll a and phycocyanin synthesis, and proline incorporation into protein is consistent with the proposed consequences of l -azetidine-2-carboxylic acid substitution for proline on the biological activity of proteins, and that the homologue is incorporated into protein intact, in addition to being converted to other intermediates, in cells of C. caldarium." @default.
