One of these has been identified as 23-keto-25-hydroxyvitamin D 3 by absorbance spectroscopy and mass spectrometry. 23-Keto-25-hydroxyvitamin D 3 was converted into a more polar metabolite by kidney homogenates prepared from vitamin D-deficient chicks. This metabolite has been identified as 23-keto-1,25-dihydroxyvitamin D 3 by mass spectrometry. 23-Keto-25-hydroxyvitamin D 3 has 4-fold higher affinity than 25-hydroxyvitamin D 3 for the tissue 1,25-dihydroxyvitamin D 3 -specific receptor. Thus, it is the first vitamin D metabolite, without a 1-hydroxyl group, that has a higher affinity for this protein than does 25-hydroxyvitamin D 3 . 1-Hydroxylation of 23-keto-25-hydroxyvitamin D 3 increased its binding potency 20-fold, producing a compound 50% as active as 1,25-dihydroxyvitamin D 3 itself.
