- W2077549797 abstract "248 Objectives: In chronic rejection of transplanted hearts, or cardiac allograft vasculopathy (CAV), early T lymphocyte and macrophage graft infiltration contributes to vascular intimal thickening. However, the mediators of T lymphocyte and macrophage recruitment in CAV are unknown. The chemokines RANTES and interferon-inducible protein 10 (IP-10) recruit T lymphocytes and monocytes in vitro. Therefore, we sought to determine the role of RANTES and IP-10 in T lymphocyte and macrophage recruitment in a vascularized murine model of CAV. Methods: B10.BR mice received a heterotopically transplanted heart from B10.A or B10.BR donors. No immunosuppression was given. Mice were sacrificed at 0,1,7,14, and 30 days after transplant. Intragraft RANTES and IP-10 expression was determined with ribonuclease protection assay. Perivascular T lymphocyte and macrophage infiltration was defined immunohistochemically and graded (0=absent, 4=diffuse, intense). Intimal proliferation was quantitated with computer-based morphometry. Results: In allografts, RANTES and IP-10 expression increased significantly at 7 days post-tranplant. This correlated with perivascular macrophage and T cell recruitment. At 14 and 30 days post-transplant, chemokine expression and cellular infiltration continued while vascular intimal thickening increased (see table). Isografts had no chemokine expression, cellular infiltrate, or intimal thickening.TableConclusions: In allografts, RANTES and IP-10 expression occurs early and correlates with perivascular macrophage and T lymphocyte accumulation. Intragraft chemokine expression and cellular infiltration persist during vascular lesion development. These results support a role for RANTES and IP-10 in macrophage and T lymphocyte recruitment during CAV development. Modulating intragraft chemokine expression may be useful in reducing cellular infiltrate and attenuating CAV." @default.
