- W2077694872 abstract "Intrauterine insemination (IUI) with donor semen is an important option for many patients seeking infertility treatment. While a simple procedure, these IUIs are most commonly offered in ART centers due to a perceived need for laboratory support and standardized conditions. However, with only around 450 infertility programs in the country, many patients do not have ready access to an infertility specialist for this type of entry level infertility treatment. The present study presents a novel semen thawing device (NSTD) which provides a highly controlled thawing environment with minimal input from an operator which would allow generalist to perform donor IUI procedures in a controlled and successful fashion. Prospective animal model study. The NSTD uses an exothermal reaction to produce a stable thawing environment. Initial studies where preformed to establish the thawing curve of the NSTD is similar to conventional water thawing (CWT). Bovine semen samples were then frozen using a controlled rate freezer and standard protocols for a thaw study. Frozen samples where thawed using either NSTD and CWT and semen parameters recorded at 0 and 3 hours post-thaw. The NSTD maintained ideal temperatures for approximately 1hr without an external energy source. Curves for both methods where equal to just above freezing (p = .826) however, above 0°C the NSTD warmed more slowly (p < .007). CWT showed higher post thaw motility and rapid cells than NSTD (p < .001), but both thawing methods produced results acceptable for IUI. Even though CWT samples had higher post thaw motility, the NSTD produced a stable thaw environment and consistent thawing rates. Additional studies are underway to improve the thawing curve observed the NSTD, additionally studies are planned to evaluate sperm biochemistry and pregnancy rates as well as studies to adapt the system to vials." @default.
