Ribosomes high in polymeric species (80 % to 85 %) and reflecting more closely the situation in vivo were obtained from green Zea mays shoots relatively high in ribonuclease using high pH (8.5) isolation media. With more mature tissue, bentonite had to be added to the homogenizing buffer to preserve the larger polysomes. The polysomes were relatively stable when incubated at 30 °C. 2. Polysomes isolated at pH 8.5, although higher in polymers, were unexpectedly no better than those isolated at pH 7.5 in in vitro amino acid incorporation. Supernatant factors obtained at pH 8.5 were more active than those obtained at pH 7.5, suggesting that the high pH dissociated some essential factors of protein synthesis from the polysomes. Nevertheless, high pH media appear to be effective for obtaining functional polysomes with minimal degradation from ribonuclease-rich tissue." @default.
