The mitochondrial configuration depends on a number of factors including the osmolarity of the isolation medium, the composition of the suspending medium, the temperature, and the time of incubation. The transition from one configuration to another involves changes in both the volume of the whole mitochondrion and in the mode of organization of the inner membrane. Mitochondria isolated in 0.44 M sucrose appear as elongated structures which become spherical when transferred into 0.25 M sucrose. This change is osmotically irreversible. Mitochondria either isolated or placed in 0.25 M sucrose appear as spheres of comparable diameter. Suspended in sucrose, they behave like perfect osmometers within a certain range of osmotic pressure. By varying the osmotic pressure of the suspending medium, the mitochondrial volume varies inversely with the osmotic pressure, provided a nonpermeant solute is used as the suspending medium. It was found that there is an almost linear relationship between volume and reciprocal of the osmotic pressure in the range between 0.22 M and 0.75 M sucrose. Data regarding the so-called “osmotic dead space” are reported. It was also observed that the thickness of the outer membrane varies inversely with the volume of the whole mitochondrion. When the outer membrane is disrupted as a consequence of a large swelling, the inner sac is extruded and appears as a large, flattened vesicle.
