- W2078507229 abstract "RATIONALE: Eosinophils derives from CD34+ bone marrow progenitors and increasing evidence suggests that these progenitors are recruited to the airways during an allergic inflammatory response. Since CC chemokine receptor 3 (CCR3) is a major receptor for eosinophil recruitment, we wanted to evaluate whether CCR3 has a regulatory effect on the recruitment of CD34+eosinophils and CD34+/Sca-1+cells in allergen-induced airway inflammation. METHODS: OVA-sensitized/exposed Balb/c mice were pre-treated intraperitoneally (i.e. systemically) with an anti-mouse CCR3 monoclonal antibody (300μg). Bronchoalveolar lavage (BAL) was collected 24 hours after the final allergen exposure. Differential cell counts and immunocytochemical detection of CD34+eosinophils and CD34+/Sca-1+cells were performed. RESULTS: Anti-CCR3 treatment significantly decreased the number of CD34+ eosinophils and CD34+/Sca-1+cells in BAL fluid (7±3 vs. 71±6 x104/ml and 7±2 vs. 55±16 ×104/ml; P<0.01) compared to isotype controls. In addition, systemic administration of anti-CCR3 reduced the number of CD34+ airway eosinophils to levels observed in mice not being allergen exposed. CONCLUSIONS: The recruitment of CD34+eosinophils into the airways in allergic airway inflammation is dependent upon the CCR3/Eotaxin pathway. Furthermore, targeting CCR3 may be effective in reducing progenitor cell mobilization in allergen-induced airway eosinophilia." @default.
