- W2078553058 abstract "A reverse hemolytic plaque assay (RHPA) was used to detect individual cells producing prosta-glandins of the E series. Epithelial cells derived from the rest of Malassez were cultured from porcine periodontal ligament on glass slides. They were incubated by the monolayer technique in liquid medium with target erythrocytes previously coated with anti-PGE2 antibodies. After 5–6 h incubation at 37 °C in the presence of complement, hemolysis plaques formed over 95% of the cells when their density was 36 cells/1.6 mm3. The percentage of plaque-forming cells was inversely proportional to the cell density of the cultures. In the absence of complement only 0–3% of the epithelial cells formed plaques. Either adsorption of the antibody with PGE2 or the addition of indomethacin (2 × 10−6 M) inhibited plaque formation. These data illustrate the usefulness of the RHPA as a general in vitro assay for detecting antigen-secreting single cells." @default.
