METHODS RCC cells in NCI-60 cancer cell panel (786O, A490, ACHN, Caki-1, RXF393, SN12C, TK10 and UO31) were used to evaluate cell death induced by HDACI (belinostat and vorinostat) and their combination with sorafenib. Cell viability was measured by CellTiter 96® Aqueous One Solution Cell proliferation assay (MTS assay) and expression of protein was analyzed by Western blot. Combination indexes were calculated with CalcuSyn software. RESULTS Treatment with belinostat or vorinostat alone inhibited growth of 8 kinds of RCC cells in NCI-60 cancer cell panel, showing several micromolar concentrations of IC50s. These HDACIs increased the acetylation of histone 3 (H3) and tubulin, and activated caspase-8, -9, and -3 in Caki-1, 786O, SN12C, and UO31 RCC cells, indicating the induction of apoptosis. Pre-treatment of clear cell type RCC (CCRCC) Caki-1 and 786O cells with HDACIs followed by sorafenib showed synergistic anti-tumor effect, activation of caspases, and decrease in Mcl-1 and Bcl-xL, phopho-Erk, and VEGF in Caki-1 and 786O cells.
