- W2078697191 abstract "Hepatocyte survival and function for prolonged period of time are limiting factors for the success of the bioartificial liver. The goal of the study was to investigate whether a new bioreactor containing Acellular Autologus Biomatrix (ABM) cultured with pig hepatocytes could prolong cell function and survival. ABM was obtained from a pig liver by immersion of liver slices in sodium desossicolate solution and DNAsi. Hepatocytes were isolated with collagenase using a standard technique. ABM and hepatocytes were placed into a bioreactor, which was continuously oxygenated. ABM and hepatocytes were perfused at 37°C for 12 and 18 h. with serum obtained from anhepatic pig. The control groups consisted of normal serum and anhepatic pig serum circulating through a bioreactor not containing hepatocytes and ABM. At 12 h. and 18 h. same results were found. Ammonia declined from 500 ± 300μg/dl to ± 70μg/dl, AST from 6000±4000 IU/L to 3000±2000 IU/L, ALT from 7000±2000 IU/L to 3000±1000 IU/L. MEGx (lidocaine 300 mg/ml) 150 mg/ml, pO2 range 200–300 mmHg, Lactid Acid was +5% respect to the untreated anhepatic serum. The control group contained AST and ALT −90%, Urea −75%, undetectable lactid acid and MEGx. Amonia was +250% respect to the normal serum. After 18 h. of perfusion, MTT and glycogen content were preserved; 100% adhesion of the cells to ABM was found. In conclusion the hepatocytes and ABM complex is able to maintain hepatocytes function and viability for prolonged period of time and guarantees 100% of hepatocytes adhesion for 18 h of continuous treatment." @default.
