METHODS The MKP-1 expression was analyzed by quantitative PCR, Western blotting and immunostainig in 51 cases of human RCCs and 2 renal cancer cell lines, i.e. A498, and 786O. The effects of human recombinant EGF (hrEGF) on the expression of MKP-1 on renal cancer cell lines were analyzed by Western blotting. Chi-square tests were performed to test the relationships between MKP-1 expression status and clinicopathological parameters. MKP-1 expression and clinicopathological variables were evaluated for association with overall survival using Kaplan-Meier survival analysis with log-rank statistic for determining significance. RESULTS When the cells were treated with hrEGF, the MKP-1 expression was up-regulated with the activation of extracellular signal-regulated protein kinase (ERK) in both cell lines. MKP-1 immunostaining was widely distributed both in tumor and matched normal kidney tissues. In normal kidney, MKP-1 expression was detected in the renal tubular epithelium. Generally, immunostaining was found in the nucleus and in the cytoplasm. RCC tumor specimens exhibited marked variability in their levels of MKP-1. Generally, immunostaining was found both in the nucleus and on the cell membrane. MKP-1 expression was up-regulated in RCCs compared with normal kidney tissue. Association between MKP-1 expression and clinicopathological variables were evaluated using chi-square test. Low MKP-1 expression was correlated with high tumor grade (p=0.0093). Survival analyses showed that low MKP-1 protein expression (p=0.0006) high tumor grade (p=0.0007), and high T stage (p<0.0001) are associated with poor overall survival. CONCLUSIONS Our cell line study results suggest the mitogenic signal, which result in ERK activation, seemed to be associated with MKP-1 up-regulation. Our data also indicated that MKP-1 expression is generally up-regulated in RCCs but is down-regulated in high grade cancer. Since low MKP-1 expression in RCCs was associated with poor overall survival, MKP-1 seems to have the potential as a prognostic marker in RCCs.
