- W2079193889 abstract "When serum bile acids are to be analysed by gas-liquid chromatography, a preliminary extraction step is necessary to remove serum proteins and other interfering substances. In the past, liquid-solid extraction with the resins Amberlyst A-26, Amberlite XAD-2 or XAD-7 has been extensively used [l-3]. More recently, the use of cartridges packed with octadecylsilane-bonded silica has been recommended for efficient and rapid extraction of bile acids and their conjugates from serum [4,5]. Sera are diluted and heated in sodium hydroxide to release bile acids bound to albumin and then passed through a cartridge containing octad~ylsil~e-bonded silica. After washing the bonded phase with water, bile acids are recovered in a small volume of methanol [4]. In the evaluation of two commercially available cartridges for extracting bile acids from serum, a problem was encountered during gas-liquid chromatography with a contaminant, which was derived from one type of cartridge. The purpose of this report is to warn laboratory workers who use reverse-phase octadecylsilanebonded silica cartridges for serum bile acid extraction that commercially available cartridges vary in their stability to alkali. As a result of the alkaline conditions under which bile acids must be extracted from serum, a persistent contaminant can be eluted from Sep-Pak-C,, cartridges along with the bile acids. This conta~nant can interfere with the subsequent separation and quantitation of bile acids by gas-liquid chromatography if bile acids are not carefully further purified prior to analysis." @default.
