• W2079586171 abstract "Over the last decade, although a number of studies have explored the relationships of APOE genotype and education with dementia, data concerning the possible modifying effect of education on the APOE - dementia association is still limited. The current study aimed to verify the hypothesis that education may decrease the risk effect of APOE ∊4 on dementia. Data were gathered from three major population based studies in Northern Europe: the Kungsholmen project, the Göteborg Birth Cohort, and the CAIDE project from Finland. Among the 3436 participants aged 65 and above, dementia was diagnosed in 219 persons according to standard criteria. Logistic regression was used to estimate the Odds Ratios (OR) and 95% confidence intervals (CI) for dementia in relation to education and APOE genotypes. APOE ∊4 was associated with increased risk of dementia independent of the effect of education (OR = 2.5 (95% CI: 1.9-3.4) and OR = 3.7 (1.8-7.2) in one and two e4 carriers, respectively). Highly educated (8 years and above) people had an OR for dementia equal to 0.5 (0.3-0.6). A biological interaction between education and APOE ∊4 was observed: as compared to less educated no APOE ∊4 carriers, persons who carry any APOE ∊4 allele had an OR of 2.7 (1.9-3.9) if they were low educated, but their OR decreased to 1.2 (0.7-1.8) if they had more than 7 years of schooling. This study suggests that genetic (APOE ∊4) and environmental (education) factors are not only independently but also interactively related to dementia risk, and that high education may buffer the negative effect of APOE ∊4 on dementia occurrence." @default.
