- W2079741427 abstract "Abstract Variants of Coxsackie A9 and ECHO 13 viruses which require 2-(α-hydroxybenzyl)-benzimidazole (HBB) for replication have been isolated and characterized. Studies with the HBB-dependent variant of Coxsackie A9 virus have shown that infective RNA extracted from the HBB-dependent virus is unable to induce production of detectable amounts of dependent virus in the absence of compound, but is able to do so in its presence. HBB-dependent virus requires the compound for its intracellular multiplication. Single-cycle growth characteristics of the dependent virus growing in the presence of an optimal concentration of HBB (22 μg/ml) were similar to those of the HBB-sensitive parent population growing in the absence of the compound. Maximal growth of the HBB-dependent variant of Coxsackie A9 virus was also obtained with 2-(o-hydroxybenzyl)-benzimidazole and 5-methyl-2- d -ribobenzimidazole, as well as with unsubstituted benzimidazole. In addition, guanidine·HCl supported maximal growth of the HBB-dependent variant. Imidazole, adenosine, arginine, histidine, and creatinine were ineffective. HBB-dependent virus which had been passaged once in the presence of 5-methyl-2- d -ribobenzimidazole or guanidine remained HBB-dependent. The HBB-dependent variant of ECHO 13 virus required somewhat higher concentrations of HBB or guanidine for maximum enhancement than did HBB-dependent Coxsackie A9 virus. Even after repeated cloning, the HBB-dependent Coxsackie A9 variant gave rise to populations containing 0.2 to 1% HBB-independent particles. Back mutation was either to HBB resistance or HBB sensitivity. The back mutation rate of ECHO 13 was less than that of Coxsackie A9. The results obtained provide evidence for the occurrence of genetically determined drug dependence among viruses." @default.
