- W2080254694 abstract "Intact lenses and isolated lens capsules from 19-day embryonic chicks synthesize hydroxy[3H]-proline-containing polypeptides when incubated in vitro with [3H]proline. The polypeptides have an apparent molecular weight of approx. 180 000 and are deposited in the deoxycholate-insoluble basement-membrane matrix. SDS/agarose gel filtration chromatography under reducing conditions indicates that with time these polypeptides are converted into aggregates of higher molecular weight. Formation of these aggregates is inhibited in the presence of β-aminopropionitrile. SDS/polyacrylamide gel electrophoresis and fluorography resolve the material of molecular weight 180 000 into three polypeptides, the largest of which has been shown to contain 4-hydroxy[3H]proline and is therefore collagenous. All three polypeptides become labelled when isolated lens capsules are incubated with [3H]fucose. The electrophoretic mobilities of the three polypeptides may be correlated with three Coomassie Blue- and periodic acid/Schiff-positive components which can be extracted from the lens capsule. These results suggest that the assembly of the embryonic chick lens capsule follows a similar biosynthetic pathway to that of the rat lens capsule although additional macromolecular components, which may represent non-collagenous glycoproteins, are present in the chick lens capsule." @default.
