Weiterhin machte er eine der glücklichsten Entscheidungen in der Geschichte der Wissenschaft: Er orientierte in der Projektionsformel der Glucarsäure die Hydroxylgruppe eines Kohlenstoffatoms nach rechts und definierte diese Konfiguration als Bezugspunkt der D/L-Klassifizierung. Erst 1951 konnte bewiesen werden, dass er die richtige Seite geraten hatte. Aus allen Puzzlesteinen wird nach Fischers Ideen die Struktur der Glucose bestimmt. On the occasion of his 150th birthday Fischer's proof of glucose's structure is shown here in the form of a stereochemical puzzle. Four of glucose's six carbons are stereogenic, thus making eight pairs of enantiomers. Fischers elucidated the structure of glucose step-by-step by using three chemical reactions: oxidation of aldoses to aldaric acids and study of their optical activities, condensation of aldoses with phenylhydrazine forming osazones, and the Kiliani reaction by which pentoses can be converted into hexoses. In addition Fischer made one of the most fortunate guesses in scientific history. He placed one of the hydroxyl groups in glucaric acids projection formula to the right, using it as a reference for the D/L classification system. Much later it was proven in 1951 that he had made the right choice. All the pieces of this stereochemical puzzle when put together according to Fischer's ideas have determined the structure of glucose." @default.
