- W2080494579 abstract "Abstract Primary T cell responses to viral and other infections are guided by factors acting at each stage of the host response, during the initial inflammatory reaction to infection, during T cell priming in the regional lymph nodes (LN), and after recruitment of activated T cells to the site of infection. The factors that shape the functional phenotype of activated T cells in vivo include the strength and character of signals delivered by antigen-presenting cells and other cells in the lymph nodes that in turn reflect the nature of the pathogen and the tissue from which they have migrated. It has not been clear, however, whether effector T cells are programmed entirely in the lymph nodes or whether they could differentiate further following recruitment to the effector site. When mice were infected with influenza virus, marked differences were observed in levels and types of cytokine expression by T cells from the mediastinal lymph nodes and lung, and cytolytic T lymphocytes (CTL) were only detected in the lung. PCR-based single-cell assays for perforin and granzyme B expression suggested that some CTL completed their differentiation in the lymph nodes and that their selective detection was mainly due to higher effector cell numbers in the infected lung. Some activated CD8+ T cells in the lung could nevertheless undergo further differentiation to express an altered cytokine profile, indicating that the T cell response can also be shaped after recruitment to the effector site." @default.
