The data indicate that the relationship between these two analytical methods depends on several factors including the size and amino acid composition of the analyte. These results led to the development of a hypothesis which predicts that the ratio of fluorescence to absorbance of the OPA-derivatives of proteolysis products can be used to identify the mode of hydrolysis. A number of proteinase enzymes of known specificity were used with a variety of protein substrates to substantiate this hypothesis. The data show that the mode of proteolysis was consistently identified; endoproteolysis results in ratios of ~ 3 while exoproteolysis shows ratios of ~ 15. Mixtures of exo- and endoproteinases result in rations of ~ 8 under the conditions employed in this work." @default.
