- W2080705492 abstract "P-157 Abstract: Supression of the immune response in occupational exposure to organic solvents is an issue controversial in immunotoxicology. In the last years there is an increasing number of reports describing various immunotoxicity effects in occupationally exposed to organic solvents. Organic solvents–induced damage to the immune system may be associated with a wide spectrum of diverse pathological conditions, some of which may be become detectable only after a long latency. In chronic exposure to benzene, macrophages may play an important role in immunotoxicity development by release of various mediators. Among them, interleukin-1 (IL-1), tumor necrosis factor (TNF) and radical oxygen species (ROS) are involved in various toxicological processes. The aim of this study was to show the effects of the chronic exposure to benzene and to mixtures of the benzene and toluene on the immune and oxidative response. In vivo experiment was carried out on Wistar rats that were divided in 8 groups as following: 1) Control-group, without exposure, sacrificed at 3 months; 2) Control-group, without exposure, sacrificed at 9 months; 3) Benzene – exposed group (500 ppm), sacrificed at 3 months; 4) Benzene – exposed group (500 ppm), sacrificed at 9 months. 5) Toluene – exposed group (800 ppm), sacrificed at 3 months; 6) Toluene – exposed group (800 ppm), sacrificed at 9 months. Benzene + Toluene – exposed group, sacrificed at 3 months; 8) Benzene+ Toluene – exposed group sacrificed at 9 months. The following parameters were assessed: a) 3HTdR incorporation test; b) IL-1 assay; c) TNF-assay; d) Chemiluminiscence assay; e) Lipid peroxides. Our results point out an important suppression of the immune response and highly activity of the cytokines, in all the exposed groups. The most important modifications of the immune and oxidative reactions have been showed in the exposed groups to benzene; in the toluene-exposed groups the modifications of the parameter shave been reduced, and in the benzene + toluene-exposed groups values of the parameters were reduced in comparison with benzene group and increased in comparison with the toluene group. The attenuated values of the investigated parameters in the benzene + toluene-exposed groups suggest a protective role of the toluene in association with benzene." @default.
