- W2080919493 abstract "175 The effects of rapamycin, an immunosuppressive drug, mirror some of the effects of the pleiotropic cytokine transforming growth factor-β (TGF-β). Both inhibit the response of activated T cells to IL-2 and other T cell growth factors, inhibit IL-1-induced interferon-γ expression in YAC cells, skew immune activation towards a Th2 response, and reduce Bcl-2 expression. We have begun to investigate whether the immunosuppressive effects of rapamycin are due, at least in part, to TGF-β induction. Splenocytes harvested from Balb/c mice and stimulated in vitro with anti-CD3 mAb and rapamycin for 72h produced more TGF-β than controls as analyzed by ELISA, although the difference was not significant. For in vivo studies, Balb/c mice were injected with allogeneic C57Bl/6 splenocytes, and subsequently treated with a single ip injection of 0, 8 or 20 mg/kg rapamycin 24 hours later. Fourteen days post-rapamycin treatment, spleens and mesenteric lymph nodes were harvested, restimulated in vitro with anti-CD3 mAb, and TGF-β production was analyzed by ELISA at 0, 24, 48 and 72h post anti-CD3 stimulation. We found that lymphocytes derived from rapamycin-treated animals produced significantly more TGF-β at 48 and 72 hours (p=0.05 by Mann-Whitney U test) than cells from vehicle-treated animals. The effect of rapamycin on TGF-β production was dose-dependent. The observation that rapamycin induces TGF-β production suggests a possible role for TGF-β in some of rapamycin's immunosuppressive effects. The effect of anti-TGF-β mAb treatment in these systems is now being explored." @default.
