- W2081036016 abstract "Abstract Two enzyme components of bee-venom—hyaluronidase and phospholipase—were separated from a third (lytic) component by ion-exchange chromatography on carboxymethyl Sephadex. The two enzymes could then be separated from each other by a previously described method of chromatography on alumina. This allowed investigation of the interactions between human serum and either crude bee-venom or its individual components. Precipitation lines appear when human serum and bee-venom diffuse towards one another in agar gel. These precipitation lines are not due to an immune reaction, since (i) the reactions were given as strongly by the sera of normal individuals as by those of bee-keepers ; (ii) the serum proteins reacting with the bee-venom were not immunoglobulins but lipoproteins ; and (iii) interactions occurred only with the phospholipase component of the venom. The essential nature of the reactions was established when it was shown that phospholipase isolated from bee-venom interacted with purified human α- and β-lipoproteins. No evidence was found of true precipitating antibodies directed against any of the components of bee-venom, even in the sera of bee-keepers who had recently suffered repeated bee-stings, but neutralizing activity against the hyaluronidase component was demonstrable in the γ-globulin fraction of bee-keepers' sera.
